Si60 TLC Plates: The “Gold Standard“ for Laboratory Separation
In the world of thin-layer chromatography (TLC), Si60 TLC plates are an unavoidable name. Since Merck introduced the classic silica gel 60 series, it has become one of the most widely used normal?phase TLC plates in laboratories worldwide, thanks to its stable performance and broad applicability. For most practitioners in organic synthesis, natural product chemistry, and pharmaceutical analysis, Si60 is virtually synonymous with TLC.
I. Technical Significance of Si60
The "60" in Si60 refers to the silica gel's average pore size of 60 ? (approximately 6 nm). This pore size determines that it is most suitable for separating small?molecule compounds with molecular weights typically below 1000 Da. For the vast majority of organic synthesis intermediates and natural products, the 60 ? pore size provides sufficient specific surface area (approximately 480–540 m2/g) and pore volume (approximately 0.74–0.84 ml/g), ensuring good sample loading capacity and separation efficiency.
The particle size of the Si60 TLC plate packing is typically 10–12 μm. The coating thickness varies depending on the backing material: glass plates generally have a coating thickness of 250 μm, while aluminium foil and plastic plates have 200 μm. This difference in thickness arises from the different adhesion requirements of the coatings on various supports, but it does not affect the core separation performance.
II. Backing Materials and Selection
Si60 TLC plates are available with several backing options to suit different experimental scenarios:

The introduction of aluminium foil and plastic plates has largely solved the problems of difficult transport, storage, and cutting associated with glass plates, significantly reducing the overall cost of using TLC plates.
III. Fluorescent Indicators: F254 and F254s
For the detection of colourless compounds, Si60 plates usually contain a fluorescent indicator. The commonly used version is Si60 F254.
· F254: The plate contains a substance that emits green fluorescence upon excitation at 254 nm UV light. When a compound in the sample absorbs UV light at this wavelength, it appears as a dark spot against the bright fluorescent background.
· F254s: Similar to F254, but its fluorescent indicator is more acid?stable, making it suitable for systems that require acidic mobile phases.
Approximately 80% of TLC applications can be established using normal?phase silica gel plates, covering alkaloids, carbohydrates, fatty acids, pesticides, steroids, tetracyclines, and many other substances.
IV. Standards and Performance
The reason Si60 TLC plates are called the "gold standard" is closely tied to their stringent performance specifications. Each batch is subjected to chromatographic testing with standard reference compounds to ensure batch?to?batch reproducibility. For example, in a typical lipophilicity visualization test, the hR<sub>f</sub> values of standard test substances must fall within specified ranges (e.g., hR<sub>f</sub> of Ceres Black G = 34–48).
In addition, Si60 TLC plates employ a polymeric binder that gives the coating a hard, smooth surface that is resistant to cracking or blistering, and even allows writing on the surface with a pencil without damaging the coating.
V. Typical Application Examples
Si60 TLC plates are extremely versatile. In natural product research, they are used for compositional analysis of plant extracts. For instance, in a study on an extract of red raspberry (S. rubriflora), researchers used 10 × 20 cm TLC Si60 F254 glass plates with ethyl acetate:methanol:water (70:20:10) as the mobile phase to achieve good separation of polar and non?polar components in the extract.
In organic synthesis, it is virtually the standard tool for reaction monitoring. Synthetic chemists commonly use aluminium?backed Si60 F254 plates to quickly assess reaction progress and product R<sub>f</sub> values by UV irradiation or staining with potassium permanganate.
